PimCell® microfluidic chips are a family of easy-to-use microfluidic devices specifically developed to handle the requirements of larger tissues in high resolution imaging experiments and assays such as confocal microscopy. Each concept supports an open top cell culture chamber connected to at least one enclosed microfluidic channel without any physical barrier between. PimCell® membrane-free technology utilizes an inverted capillary pressure barrier (CPB) between an open-top chamber and a precisely aligned microfluidic channel, ensuring a clear, unobstructed visualization between the chamber and the channel. When ECM is added to the open-top chamber, it forms a meniscus directly on the glass bottom of the adjacent microfluidic channel. Large compartments of PimCell® devices improve long-term culture and are especially useful for culturing human stem cell-derived in vitro models, providing better long-term growth. Our devices can be used with gravity driven perfusion, supporting in vitro reconstruction of 3D tissue microenvironment. The PimCell® family is designed as an alternative to existing microfluidic devices which have relative large space without sufficient perfusion inside hydrogel-filled microfluidic channels. The open top cell culture chamber eliminates the need of a hydrogel introduction channel with its dead volume. Any desired 3D cell culture support can be placed directly near the microfluidic channel. The direct advantage is an opportunity to create a novel or to upgrade existing in vitro models for producing more valuable and reliable data.
